چکیده
OBJECTIVE To evaluate the recently available Cobas Amplicor polymerase chain reaction (PCR) system for the detection of Mycobacterium tuberculosis complex (MTBC) in well-characterized clinical specimens and to compare the results with clinical classification, conventional culture and staining techniques. METHODS Three hundred and forty-four clinical specimens consecutively received for culture of acid-fast bacilli by the laboratory of Zayed Military Hospital, Abu Dhabi, United Arab Emirates, from 2004 to 2005, were used in this study. Acid-fast bacilli and culture for tuberculosis were compared with Cobas Amplicor PCR. RESULTS The final diagnostic evaluation of the Cobas Amplicor MTB showed the followings: sensitivity 73.3%, specificity 99.4%, positive predictive value 84.6%, negative predictive value 98.8% and with accuracy of 98.2% for the Cobas Amplicor MTB assay. CONCLUSION The automated Cobas Amplicor MTB assay is suitable for routine use in clinical laboratories.

Detection of Mycobacterium tuberculosis complex in sputum specimens by the automated Roche Cobas Amplicor Mycobacterium Tuberculosis Test.
Three hundred twenty-four sputum specimens from 151 patients with suspected active pulmonary tuberculosis were tested for the presence of the Mycobacterium tuberculosis complex with auramine fluorochrome stain and automated PCR assay (Roche Cobas Amplicor Mycobacterium Tuberculosis Test [MTB]). The results were compared with those of the conventional Löwenstein-Jensen tube culture and the BACTE...
متن کاملEvaluation of Cobas TaqMan MTB for direct detection of the Mycobacterium tuberculosis complex in comparison with Cobas Amplicor MTB.
The Roche Cobas Amplicor MTB assay, recently replaced by the Roche Cobas TaqMan MTB assay, was one of the first commercially available assays for detection of the Mycobacterium tuberculosis complex based on nucleic acid amplification. We reported previously on the limited specificity of the Cobas Amplicor MTB assay, in particular for positive samples with an optical density at 660 nm (OD660) of...

PURPOSE Polymerase chain reaction (PCR) assay, introduced as a fast and sensitive diagnostic method, is useful in detecting Mycobacterium tuberculosis. The purpose of this study was to evaluate the usefulness of in-house PCR assay in the detection of Mycobacterium tuberculosis by comparing PCR results with conventional diagnostic techniques and Cobas Amplicor M. tuberculosis kit. MATERIALS AN...
متن کاملEvaluation of Cobas TaqMan MTB PCR for detection of Mycobacterium tuberculosis.
Nucleic acid-based amplification tests allow the rapid detection of Mycobacterium tuberculosis. Recently, a real-time PCR assay for M. tuberculosis complex, the Cobas TaqMan MTB test (Roche Diagnostics, Basel, Switzerland), was introduced. We performed a prospective study to evaluate the diagnostic performance of the Cobas TaqMan MTB test system. A total of 406 specimens collected from 247 pati...
متن کاملPotential for the Use of Polymerase Chain Reaction (PCR) in the Detection and Identification of Mycobacterium tuberculosis Complex in Sputum Samples
The COBAS AMPLICOR Mycobacterium tuberculosis assay was evaluated for routine detection and identification of Mycobacterium tuberculosis complex (MTBC) in 629 sputum samples. In addition to the Polymerase Chain Reaction (PCR), diagnostic culture, which is considered the reference method, was also performed on theses samples. Of the 629 specimens tested, 64 were culture positive for M. tuberculo...
